Structure

CJC-1295 No DAC is the first 29 residues of human GHRH (the shortest fully active fragment, also known as sermorelin) with four substitutions that make it more resistant to enzymatic breakdown: D-Ala at position 2, Gln at 8, Ala at 15 and Leu at 27. The C-terminus is amidated.

“No DAC” means it lacks the Drug Affinity Complex, a maleimidopropionyl-lysine extension that lets the related compound CJC-1295 with DAC bind covalently to albumin. The two are different molecules: CJC-1295 with DAC is ≈3,647 g/mol and carries its own CAS number (446262-90-4).

Ipamorelin is a five-residue peptide with three non-standard building blocks: α-aminoisobutyric acid (Aib), D-2-naphthylalanine (D-2Nal) and D-phenylalanine, plus a C-terminal amide. It was developed at Novo Nordisk as a ghrelin-receptor agonist.

Mechanisms studied in research models

  • Two separate receptors. GHRH-receptor agonists and ghrelin-receptor agonists act through different pathways in pituitary somatotrophs (Gs/cAMP for the GHRH receptor; Gq/calcium for GHSR-1a). Pairing them lets researchers study how the two inputs combine.
  • Ipamorelin selectivity. In the discovery study, ipamorelin released GH from rat pituitary cells in vitro. In swine, it raised GH without raising ACTH or cortisol above control at exposures far above those needed for GH release, unlike the older secretagogues GHRP-6 and GHRP-2 (Raun et al., 1998).
  • Limited primary literature for Mod GRF 1-29. Most published CJC-1295 studies use the DAC version. The tetrasubstituted analogue without DAC has little dedicated primary literature, so its properties are mostly inferred from GHRH(1-29) structure–activity work.
