Selank (also called TP-7) is a synthetic heptapeptide, Thr-Lys-Pro-Arg-Pro-Gly-Pro. It joins the natural tetrapeptide tuftsin (Thr-Lys-Pro-Arg) to a Pro-Gly-Pro tail that slows breakdown by peptidases. It was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences. Laboratories use it to study expression of GABA-system genes in rodent brain and in neuronal cell lines. Structure
Selank has two parts. The first four residues, Thr-Lys-Pro-Arg, are tuftsin, a peptide naturally released from the Fc region of immunoglobulin G. The last three, Pro-Gly-Pro, are a proline-rich extension used in several Russian-developed peptides (including Semax) to resist enzymatic degradation. The result is a short linear peptide with no modifications at either end.
Mechanisms studied in research models
- GABA-system gene expression in brain. In rats, Selank changed mRNA levels of several genes involved in GABAergic neurotransmission, measured by quantitative PCR (Volkova et al., 2016).
- Gene expression in neuronal cells. In IMR-32 human neuroblastoma cells, Selank, GABA and olanzapine each changed expression of GABA-pathway genes, and the patterns were compared (Filatova et al., 2017).
- Tuftsin lineage. Because it contains tuftsin, Selank is also studied in relation to immune-cell signaling, the original context in which tuftsin was characterized.
